Complement Fixation Tests in Poliomyelitis **

چکیده
Infection with poliomyelitis virus gives rise to complement-fixing (C-F) antibodies which may be detected with antigens prepared either from the brains of infected mice or tissue culture fluids, as shown with Type II by Casals et al."' and with all three types by Svedmyr et al.' These workers used sufficient antigen in each test to obtain maximum sensitivity. With this antigen concentration the presence of antibody could generally be detected within the first week after onset of the disease, but the sera usually gave similar C-F titers against antigens of the three types of poliomyelitis virus regardless of the type involved in the infection. It has now been found possible to obtain a more specific reaction in patients with the disease by a method in which not more than one conventional unit of antigen is used per test. In order to detect fixation with this low level of antigen, dilutions of serum were titrated at several complement concentrations by the plate method of Fulton and Dumbell.'

A Complement Fixation Test for Poliomyelitis
A macroscopic (tube) complement fixation test for poliomyelitis, using infected tissue culture fluids, is described. The test was applied to 27 individuals with a clinical diagnosis of poliomyelitis. In 18 patients it was possible to make a laboratory diagnosis of poliomyelitis on the basis of a rise in complement-fixing antibody titer and in 4 others on the basis of a high stationary antibody ...
متن کاملThe use of new antigens in the complementfixation test for acute poliomyelitis.
Hitherto in poliomyelitis complement-fixation tests on human sera the antigenic material has usually been derived from tissue cultures of polio viruses. Despite high infectivity titres, antigens from this source are weak in capacity to fix complement (Selzer, 1958). Concentration of tissue culture antigens improves the complement-fixing power but frequently produces anti-complementary effects. ...
متن کاملComplement fixation test using the antigen prepared from the nervous tissue of adult mice infected with poliomyelitis virus Lansing type.
The technique of complement fixation test as a measure for the serological diagnosis of neurotropic virus infections was first put in practice by Casals and Palacios (1) in the year 1941, and confirmed of its practical diagnostic value by numerous follow-ups made after it. In spite of repeated attempts, the application of this technique for the diagnosis of poliomyelitis had not been successful...
متن کاملDevelopment, Persistence, and Significance of Type 2, Poliomyelitis Complement-fixing Antibody in Man
Sera from 81 patients with a diagnosis of paralytic or non-paralytic poliomyelitis, and from 159 individuals of similar age groups giving no history of the disease, were tested with a high titered, complement-fixing poliomyelitis antigen of Type 2 (Lansing-like). The antigen consisted of brain tissue from newborn mice injected with the MEF1 strain of virus as previously adapted to these animals...
متن کاملComplement-fixing Antibodies to Type 2 (lansing) Poliomyelitis Virus in a Normal Population of a Subtropical Area
Sera collected in 1950 from the native population in the vicinity of Cairo, Egypt, have been tested for complement-fixing antibodies to Type 2 (Lansing) poliomyelitis virus. Complement-fixing antibodies are confined to the age of 1 to 9 years if a serum dilution of 1:4 is used in the test, or to the age of 1 to 4 years with a serum dilution of 1:16. A comparison has been made of the findings ob...
